Description:

Desthiobiotin PEG NHS is one of Nanocs desthiobiotin PEG derivatives that can react with primary amine groups. Desthiobiotin has similar binding properties to avidin or strepatvidin as Biotin but it is elutable from streptavidin, making it ideal for protein purification and intracellular labeling. The desthiobiotin tag binds to streptavidin and other biotin-binding proteins with high specificity yet readily elutes with mild conditions (i.e., by competitive displacement with regular, free biotin). As such, this reagent is a useful alternative to Biotin-NHS for avidin-biotin techniques in which nondenaturing elution of the labeled proteins is desired. This simplest and smallest amine-reactive desthiobiotin reagent is uncharged and cell membrane permeable; therefore, NHS-Desthiobiotin can be used with intact cells for intracellular protein labeling. N-hydroxysuccinimide ester of desthiobiotin reacts efficiently with primary amine groups (-NH2) in alkaline buffer (pH 7-9). Pegylated desthiobiotin has a hydrophilic PEG chain between NHS and desthiobitin and thus offers higher water solubility and less steric hindrance.
